How deep should sand be in tank?

No deeper than 1 inch.

1 inch of any substrate should normally suffice, but its not too big of a deal to go deeper. Most folks with planted tanks shoot for an average depth of 2-3" sometimes deeper than that towards the back, and many use sand. The only real problem with deep sand (so I've heard, never experienced) is pockets can build up in the sand of aenarobic bacteria and supposedly posienous gasses will build up and leech into the water. This can be remedied by stirring up the sand with a stick, havind snails like MTS that move through the sand constantly, and perhaps just loaches rummaging around in the sand.
